The pair had been included in the provisional list, which was announced in July, but narrowly missed the cut alongside Yusuf Abdulla, Justin Ontong and Vaughn van Jaarsveld for the September 22 to October 5 tournament, which will be played on home soil in Pretoria and Johannesburg.
Langeveldt, 34, was a surprise name in the initial squad having last represented the Proteas more than 17 months ago and having signed a two-year Kolpak contract with English county side Derbyshire in April last year.
However, according to South Africa's convenor of selectors Mike Procter his axing was due to injury, while the dropping of Morkel comes after his continued loss of form.
Procter said: "There were some difficult choices to make. At the moment South African cricket is blessed with an abundance of fast bowling talent, with the emergence of (Lonwabo) Tsotsobe and Wayne Parnell as outstanding new prospects and Charl Langeveldt becoming available again.
"Langeveldt is not fit enough yet to be considered after his shoulder surgery and Morne Morkel is the unlucky one to miss out.
"He is very much part of our long-term planning and has a major role to play but we would like to see more consistency from him at this stage.
"Planning for the 2011 World Cup is very much part of our thinking but there is a lot of cricket to be played before then. We must take our selections series by series and not get ahead of ourselves.
"The selection panel attaches great value to experience, which is well illustrated by the mentoring role Makhaya Ntini has fulfilled with Tsotsobe.
"Bringing players like Tsotsobe, Parnell and Roelof van der Merwe into a winning culture has also enabled them to mature as international cricketers quickly."
After an outstanding opening season playing in England, Langeveldt injured his shoulder following his participation in the Indian Premier League, which ruled him out for the remainder of the season.
Tsotsobe, 25, has only one cap to his name, which came in an outstanding display against Australia earlier this year when he claimed four for 50.
But a knee injury has kept him out since then, forcing him to miss the home series against the Australians as well as the ICC World Twenty20.
Procter also praised South Africa's depth across the board, saying it offered plenty of options to captain Graeme Smith and coach Mickey Arthur.
He added: "We have four quality spin bowlers and two all-rounders in Kallis and Albie Morkel to back up the seam attack. There is also batting potential most of the way down the order."
The squad will assemble in Potchefstroom at the end of the month for a six-day training camp - including two practice matches against local side Highveld Lions.
