The lunchtime kick-off sees champions Manchester United visit arch rivals Liverpool at Anfield in a blockbuster tie.
Liverpool defender Alvaro Arbeloa is desperate for his team to make amends for last season's defeat at home to the champions, when Carlos Tevez's strike two minutes before half-time condemned the Merseyside outfit to a 1-0 loss."That was a really bad experience at Anfield last season," the Spain international said to www.liverpoolfc.tv.
"I know how much of an important game it is for us and our supporters and we were all really disappointed.
"I think this game is worth much more than three points. If we beat Manchester United at home we will have 10 points and much more confidence. But everyone will also be very, very happy."
Chelsea and Liverpool top the Premier League standings on seven points, one clear of Manchester City, Arsenal, West Ham and Middlesbrough.
Brazil forward Robinho could make his Manchester City debut on Saturday against the team he spent most of the summer hoping to join.
The 24-year-old made it clear he wanted to leave Real Madrid and was widely expected to link up with compatriot Luiz Felipe Scolari at Chelsea.
He held an impromptu press conference on August 31 at which he reaffirmed: "The situation is still the same, I want to play for Chelsea as I have told the president (Ramon Calderon), the coach (Bernd Schuster), the sporting director (Predrag Mijatovic). And a twist of fate sees the first round of Premier League fixtures after the international break pit City against Chelsea.
City fans are desperate to see Britain's costliest player but the US$64 million signing is not due to return from Brazil's World Cup encounter with Bolivia until Friday lunchtime.
It means he will hardly have spent any time in England, met his team-mates or trained.
The game will also be a special occasion for Shaun Wright-Philips, facing the club where he spent three frustrating years on his first home appearance since rejoining the Eastlands outfit.
The Gunners - including England's hat-trick hero in Wednesday's impressive 4-1 victory in Croatia, Theo Walcott - travel to Blackburn on Saturday while Boro visit Portsmouth.
West Ham, who on Wednesday agreed a three-year deal with Gianfranco Zola to become the club's next manager, visit West Bromwich Albion.
Newcastle - in their first match since Kevin Keegan quit his post as manager last week, less than eight months after taking charge of the club for a second time - take on top-flight new boys Hull City.
Also on Saturday, Wigan face Sunderland and Fulham play Bolton. On Sunday, Everton visit Stoke and the fourth round of fixtures concludes on Monday night when Tottenham host Aston Villa.
